In een tweede gerandomiseerd, dubbelblind, klinisch multicenteronderzoek met parallelgroepen werd behandeling met aprepitant vergeleken met standaardtherapie bij 848 patiënten (652 vrouwen, 196 mannen) die chemotherapie kregen met een IV dosis oxaliplatine, carboplatine, epirubicine, idarubicine, ifosfamide, irinotecan, daunorubicine, doxorubicine; cyclofosfamide IV (< 1500 mg/m 2 ); of cytarabine IV (> 1 g/m 2 ).

1.060 patiënten werden gerandomiseerd naar docetaxel 75 mg/m² toegediend 1 uur na doxorubicine 50 mg/m² en cyclofosfamide 500 mg/m² (539 patiënten in de TAC-groep) of doxorubicine 50 mg/m² gevolgd door fluoro-uracil 500 mg/m² en cyclofosfamide 500 mg/m² (521 patiënten in de FAC-groep) als adjuvante behandeling van patiënten met een operabele lymfkliernegatieve borstkanker met een hoog risico op relaps volgens de Sankt Gallencriteria van 1998 (tumorgrootte > 2 cm en/of ER- en PR-negatief en/of hoge histologische nucleaire graad (graad 2 tot 3) en/of leeftijd < 35 jaar).
1060 patientes ont été randomisées à recevoir soit 75 mg/m² de docétaxel administrés 1 heure après 50 mg/m² de doxorubicine et 500 mg/m² de cyclophosphamide (539 patientes dans le bras TAC), soit 50 mg/m² de doxorubicine suivis de 500 mg/m² de fluorouracile et de 500 mg/m² de cyclophosphamide (521 patientes dans le bras FAC), comme traitement adjuvant des patientes atteintes de cancer du sein opérable avec ganglions négatifs à haut risque de rechute selon les critères de Saint-Gall 1998 (taille de la tumeur > 2 cm et/ou RO et RP négatifs et/ou grade histologique/nucléaire élevé (grade 2 à 3) et/ou âge < 35 ans.
