Een gerandomiseerd, parallelgroep, open-label, fase-III-multicenteronderzoek waarin de veiligheid en werkzaamheid van Caelyx plus bortezomib combinatietherapie werd vergeleken met bortezomib monotherapie bij patiënten met multipel myeloom die ten minste 1 eerdere behandeling hebben gekregen en die geen verbetering ondervonden terwijl ze een op antracycline gebaseerde therapie kregen, werd uitgevoerd bij 646 patiënten.
Une étude de phase III randomisée, en groupes parallèles, ouverte, multicentrique, comparant la tolérance et l'efficacité de l'association Caelyx + bortézomib versus bortézomib en monothérapie chez des patients atteints de myélome multiple qui avaient reçu au moins un traitement antérieur et qui n'avaient pas progressé pendant le traitement à base d'anthracycline, a été conduite chez 646 patients.

Patiënten met multipel myeloom: Van de 646 patiënten met multipel myeloom die ten minste 1 eerdere behandeling hebben gekregen, werden 318 patiënten behandeld met de combinatietherapie van Caelyx 30 mg/m 2 als een 1 uur durende intraveneuze infusie toegediend op dag 4 na bortezomib dat wordt toegediend in een dosis van 1,3 mg/m² op dag 1, 4, 8 en 11, om de drie weken of met bortezomib monotherapie tijdens een klinisch fase-III-onderzoek.
Myélome multiple : Dans un essai clinique de phase III, sur 646 patients atteints de myélome multiple qui ont reçu au moins un traitement antérieur, 318 patients ont été traités par l'association de Caelyx 30 mg/m 2 , en perfusion intraveineuse d'une heure administrée le 4 ème jour après le bortézomib qui est administré à 1,3 mg/m² à J1, J4, J8 et J11, toutes les trois semaines ou par le bortézomib en monothérapie.

Neutropenie, trombocytopenie en anemie waren de meest frequent gemelde hematologische bijwerkingen met zowel de combinatietherapie van Caelyx plus bortezomib als met bortezomib monotherapie.
Neutropénie, thrombocytopénie, et anémie étaient les événements hématologiques les plus fréquemment rapportés avec l'association Caelyx + bortézomib et avec le bortézomib en monothérapie.

Er was een significante verbetering in het primaire eindpunt van de tijd tot progressie (TTP) voor patiënten behandeld met de combinatietherapie van Caelyx plus bortezomib vergeleken met patiënten behandeld met de bortezomib monotherapie zoals aangetoond door een risicoreductie (RR) van 35 % (95 % CI; 21-47 %), p < 0,0001, gebaseerd op 407 TTPgevallen.
Une amélioration significative du critère principal temps avant progression (TTP) a été observée pour les patients ayant reçu l'association Caelyx + bortézomib comparativement aux patients ayant reçu le bortézomib en monothérapie, amélioration indiquée par une réduction du risque (RR) de 35 % (IC 95 % ; 21-47 %), p < 0,0001, basée sur 407 événements TTP.

De meest voorkomende (geneesmiddelgerelateerde, uit de behandeling voortkomende) bijwerkingen tijdens de combinatietherapie (Caelyx + bortezomib) waren misselijkheid (40 %), diarree (35 %), neutropenie (33 %), trombocytopenie (29 %), braken (28 %), vermoeidheid (27 %) en constipatie (22 %).
Les événements indésirables les plus fréquemment rapportés (survenus pendant le traitement et imputés à celui-ci) sous l’association Caelyx + bortézomib étaient les suivants : nausées (40 %), diarrhée (35 %), neutropénie (33 %), thrombocytopénie (29 %), vomissements (28 %), fatigue (27 %) et constipation (22 %).

PPE werd gemeld bij 16 % van de patiënten met multipel myeloom die een combinatietherapie van Caelyx plus bortezomib kregen.
